转录因子ZEB1与乙醛刺激后大鼠肝星状细胞增殖的关系分析 被引量:1

摘要 目的:探讨转录因子ZEB1与乙醛刺激后大鼠肝星状细胞增殖的关系。方法:培养大鼠肝星状细胞并将其随机分为实验组(乙醛终浓度200μmol/L)和对照组(加入等量培养基),分别培养6h,12h,24h。利用MTT法检测各时间点的各组细胞增殖情况;ELISA试剂盒检测各组细胞分泌Ⅰ型胶原蛋白含量变化;Western blotting检测细胞提取蛋白包括ZEB1、α-SMA的表达情况。结果:乙醛刺激大鼠肝星状细胞后,细胞增殖明显;Ⅰ型胶原蛋白、ZEB1及α-SMA等蛋白的表达上调,且细胞增殖程度与Ⅰ型胶原蛋白、ZEB1及α-SMA等蛋白的表达上调呈正相关。结论:乙醛刺激大鼠肝星状细胞后ZEB1通过参与大鼠肝星状细胞EMT过程,从而使细胞增殖,EMT与酒精性肝纤维化的发生发展有一定的关系。 Objective:To study the role of ZEB1 in rat hepatic stellate cell proliferation stimulated by aeetaldehype. Methods: Cultured rat hepatic stellate cells were divided into test group,treated with aeetaldehype (final eoneetration 200μmol/L ), and control group.HSC proliferation was detected by MTT,expression of collagen I by ELISA and protein level of ZEB1 and cx-SMA by Western blotting. Results: After acetaldehype stimulation, proliferation of HSC was activated ,expressions of collagen I ,ZEB1 and oL-SMA were up-regulated,and the up- regulation was positively correlated with ceil proliferation. Conclusion: After rat hepatic stellate cells were stimulated by acetaldehype,ZEB1 may participate in the process of epithelial-mesenehymal transition(EMT)to activate the cell proliferation,and EMT may be in certain relation to pathogenesis of alcoholic liver fibrosis.
