摘要
目的建立一种直接测定尿样中马尿酸含量的高效液相色谱方法。方法采用甲醇:磷酸盐缓冲液(20:80 V/V)为流动相,流速0.8 ml/min,PE4.6×150 mm C18/5μm柱分离,二极管阵列检测器检测波长220 nm,峰鉴别定性,峰面积定量。结果在选定条件下,标准曲线线性良好,相关系数大于0.999 2,方法检出限为1.2 mg/L,相对标准偏差(RSD)在0.98%~7.90%之间,回收率在91.0%~105.0%之间。结论方法简便,精密度、准确度满足要求。
Objective To establishment a direct method to determine hippuric acid in urine with High Performance Liq- uid Chromatography (HPLC). Methods Experimental settings were as following: methanol and phosphate buffer solu- tion (20:80 V/V) as mobile phase; velocity 0.8ml/min; separation column, PFA. 6×150mm C18/51μm and the di- ode array detector wavelength, 220 nm. Peak was identified qualitatively and peak area, quantitatively. Results Under the selected chromatography conditions, the linear relationship of standard curve was excellent, the correlation coefficient was above 0.9992; the detection limit was 1.2mg/L. The relative standard deviation was between 0.98% -7. 9% and the recovery rate was in 91.0% - 105.0%. Conclusion The method is proved to be simple, precise and accurate.
