A Study on in vitro Rapid Propagation Technology for Crisp Plum in Guizhou Province

摘要 以酥李带芽茎段为外植体进行组织培养,研究了植物生长调节剂组合对腋芽诱导、增殖及生根的影响,初步建立酥李离体快繁技术体系。结果表明,外植体消毒采用1 g/LHgCl2浸泡8 min的效果较好,适宜腋芽诱导培养基为WPM+1.0 mg/L 6-BA+0.5 mg/L KT+0.1 mg/L NAA,适宜芽增殖培养基为WPM+1.0 mg/LKT+0.1 mg/L IBA+1.0 mg/L GA3,适宜生根培养基为1/2MS+0.4 mg/L IBA+15 g/L蔗糖,其生根率达95%,移栽成活率在90%以上。 Using the axillary growth regulators on differentiation rapid propagation techniques were buds as explants for tissue culture, the , proliferation and rooting of shoots in Crisp established. The results showed that the effects of combinations Plum were investigated of plant , and the optimal disinfection time of ex- plants in 0.1% HgCl2 was 8 min. The optimal medium for initial culture was WPM + 1.0 mg/L 6 - BA + 0.5 mg/LKT +0.1 mg/LNAA. The optimal multiplication medium was WPM + 1.0 mg/LKT +0. lmg/L IBA + 1.0 mg/L GA3. The optimal medium for rooting was 1/2MS + 0.4 mg/LIBA + 1.5% sugar, and its rate for rooting reached 95% and rate of transplanting survival reached 90%.
