摘要
以白背三七带芽嫩叶和茎段为外植体诱导愈伤组织,建立愈伤组织培养体系,并对主要培养条件进行了优化。结果表明:白背三七带芽嫩叶在MS培养基与1.0 mg/L KT+0.5 mg/L 2,4-D+0.3 mg/L NAA+1.0 mg/L 6-BA激素组合下能快速诱导出愈伤组织,最佳诱导时期为3月份。白背三七在B5培养基与0.5 mg/L NAA+2.0 mg/L 6-BA激素组合下可获得疏松易碎、有金属光泽、生长旺盛的愈伤组织,呈平缓的"S"型生长曲线,接种后第18天生物量达到最大值19.54 g/瓶。
Leaves with buds and stem sections of Gynura divaricata ( L. ) DC. were used to induce callus. Optimum conditions and hormone combinations were constructed for callus growth and subculture. The results showed : leaves with buds of Gynura divaricata could induce callus on the culture medium of MS + 1.0 mg/ L KT + 0.5 mg/L 2,4 - D + 0.3 mg/L NAA + 1.0 mg/L 6 - BA and the best time for inducing treatment was in March. Gynura divaricata could produce crisp vigorous callus with metal luster on the Bs medium com- bined with 0.5 mg/L NAA + 2.0 mg/L 6 - BA. The growth curve was in the shape of "S", and the highest biomass yield reached 19.54 g/bottle on the 18th day after inoculation.
