摘要 目的探讨天然化合物蛇床子素对肺鳞癌NCI-H520细胞增殖及凋亡的影响。方法采用MTI比色法检测蛇床子素对NCI-H520细胞增殖的影响;流式细胞仪AnnexinV/PI双染法检测诱导细胞凋亡;Hoechst33342核染色观察细胞核的形态变化。结果蛇床子素对NCI-H520有明显的增殖抑制作用,且呈现明显时间剂量依赖性;流式细胞仪AnnexinV/PI双染法结果显示蛇床子素可以诱导细胞凋亡,且凋亡率呈剂量依赖性;药物处理组细胞核经Hoechst 33342染色可见染色质边集、核碎裂等典型的细胞凋亡改变,而空白对照组没有相应的变化。结论蛇床子素可以抑制NCI-H520细胞的增殖、诱导细胞凋亡。 Objective To study effects of osthole on cell proliferation and apoptosis in human squamous cell lung carcinoma NCI-H520 cells. Method We used the MTT assay to measure the cell proliferation. Apoptosis was determined by flow cytometry analysis by using an Annexin V-FITC Apoptosis Kit. The changes of the nucleus were investigated by Hoechst 33342 dyeing. Result The growth of NCI-H520 cells was suppressed by osthole in a dose-and time-dependent manner. Flow cytometric analysis showed that osthole induced cells apoptosis in a dose-dependent manner. Chromosome condensation and nuclear fragmentation were observed by Hoechst 33342 dyeing in treated NCI-H520 cells, the untreated cells remained normal. Conclusion Osthole could inhibit the growth of NCI-H520 cells and induce cell apoptosis.
