黑龙江省稻瘟病菌遗传多样性研究 被引量:4

Genetic diversity of Magnaporthe grisea from rice in Heilongjiang Province

摘要 利用8个代表性菌株从153对引物组合中筛选出多态性高且稳定性好的10对引物组合,对来自黑龙江省各水稻主要产区的214个稻瘟病菌菌株进行RSAP(Restriction site amplified polymorphism)分析,得到134条多态性条带,经聚丙烯酰胺凝胶电泳,得到196个不同单元型(带型)的代表性菌株的指纹图谱。用UPGMA进行聚类分析,以相似系数0.83为界,将214个菌株196个单元型划分为10个遗传宗谱,10个遗传宗谱在黑龙江省的分布存在明显地域差异。 Total 10 higher polymorphism and good stability pairs of primers were selected from 153 pairs of RSAP primers by using DNA of eight typical isolates of Magnaporthe grisea. Total 134 different polymorphism stripes were obtained after RSAP analysis of 214 Magnaporthe grisea isolates from Heilongjiang Province were done. Total 196 different haplotypes (stripes types) of the representa- tive RSAP fingerprint were aquired by polyacrylamide gel electrophoresis. 0.83 was as the similarity coefficient boundary, 214 strains were divided into 10 genetic lineages by cluster analysis using UPGMA software. Magnaporthe grisea in Heilongjiang Province showed rich genetic diversity, and the distribution of the 10 genetic lineages were obviously area difference.
