摘要 根据我国西北部胃癌发病率高的特点,通过RNA干扰技术,抑制胃癌细胞SGC7901的livin基因表达后,采用RT-PCR和细胞生长曲线法,分析胃癌细胞SGC7901的恶性生长、增殖特性.发现成功沉默livin基因后,SGC7901的生长速度明显减慢、恶性表型减轻.抑制livin基因的表达可以作为治疗胃癌的潜在靶点. Based on the characteristics of high incidence of gastric cancer in the northwest of China, inhibi- tion of livin gene expression in gastric cancer cells of SGC7901 through the RNA interference technology. RT-PCR was used to validate the gene-silencing efficiency of livin in SGC7901 cells. The cell growth rate was measured to analyze the effect of livin on SGC7901/livin siRNA cell's proliferation. It was found that the growth of SGC7901 speed significantly became slowly and malignant phenotype eased after successful si- lence livin gene. Inhibition of livin may be a potential target for the treatment of gastric cancer.
