银屑病患者外周血T细胞SCL及NEAT1的表达 被引量:3

Expressions of SCL and NEAT1 in peripheral blood T cells in patients with psoriasis

摘要 目的分析寻常性银屑病患者外周血T细胞SCL和NEAT1mRNA的表达情况。方法收集20例银屑病患者及20名健康对照外周血,分离、培养、扩增外周血T细胞并鉴定纯度;提取纯化mRNA并反转录合成cDNA,实时荧光定量聚合酶链反应(RFQ-PCR)检测SCL及NEAT1在mRNA水平的表达。结果银屑病患者外周血T细胞SCL及NEAT1基因表达水平均低于健康对照组,银屑病组与对照组SCL表达比值为0.316759,NEAT1比值为0.538544。结论 SCL表达异常可能通过其靶基因RUNX1参与了银屑病患者外周血功能性T细胞分化不平衡;NEAT1表达水平降低可能导致血红蛋白在合成、运输水平发生变化,进而影响致病菌的发育、繁殖。SCL、NEAT1通过不同的作用机制参与了银屑病的发病。 Objective To analyze the expressions of SCL and NEATI in peripheral blood T cells in patients with psoriasis vulgaris. Methods Twenty cases of psoriasis and 20 cases of control peripheral blood were selected and isolated, cultured, amplified their peripheral T cells so as to determine the degree of purity. Thereafter, the puri- fied mRNA was extracted and reversed into eDNA. The mRNA expressions of SCL and NEATI were detected by real- time fluorescent quantitative PCR (RFQ-PCR). Results The expressions of SCL and NEATI of peripheral blood T- cells in patients with psoriasis were lower than those in normal controls. The ratios of SCL and NEATI were 0.316 759 and 0.538 544 in both psoriasis and control i^oups, respectively. Conclusion The abnormal expression of SCL might be participate in the differentiation imbalance of T cells in patients with psoriasis through its target gene RUNX1. The decreased expression of NEAT1 might lead to the changes in synthesis and trafficking of heine, which further effected the development and reproduction of pathogenic bacterium. Therefore, SCL and NEAT1 might partici- pate in the occurrence of psoriasis through different mechanisms.
