放射性脑损伤兔模型的建立

摘要 目的建立放射性脑损伤兔模型,为进一步深入研究放射性脑损伤早期诊断的磁共振成像(MRI)研究提供基础。方法 CT定位下建立三维重建图像资料,通过放射治疗系统精确制定三维适形放疗计划,对2组新西兰兔分别行半脑30Gy、100Gy剂量照射。每日观察其摄食、饮水量,自主活动情况及相关体征。每周复查MRI检查,直至MRI出现放射性脑损伤或随访满16周为止。结果 30Gy组仅1例出现了硬膜下血肿,余未见明显异常。100Gy5例MRI均出现不同程度信号异常,病理证实为放射性脑病。结论通过放射治疗系统制定放疗计划,制作放射性脑损伤兔模型,能够较好地模拟放射性脑损伤过程,该模型能够适用于临床常规线圈的MRI检查。 Objective To establish the model of radiation-induced cerebral injury in rabbits for investigation on the early diagnosis by using magnetic resonance imaging (MRI). Methods By employing the stereospeclfic three-di- mensional conformal radiation therapy system, the rabbits of both groups were subjected to hemispheic irradiation with 30 Gy and 100 Gy, respectively. Ingestion of food and water, spontaneous activity and vital signs were monitored on a daily basis. The MRI was performed weekly to which definitive diagnosis of radiation-induced cerebral injury was es- tablished or a 16-week follow-up schedule was fulfilled. Results A signle rabbit showed subdural hemorrhage on MRI in the group receiving radiation of 30 Gy. Five rabbits evidenced definitive diagnosis of radiation-induced cere- bral injury on MRI featured by aberrant signals to varying extents. Conclusion By establishing the rabbit model of radiation-induced cerebral injury, the pathogenesis can be well-simulated and applied clinically to conventional MRI, thus providing a sound basis for further studies that will target at exploring the mechanisms of radiation-induced cere- bral injury.
