益气化瘀养阴解毒方药物血清对人肺癌A549细胞增殖及周期的影响

Effect of Serum Containing Decoction of Strengthening Qi, Clearing Stasis, Nourishing Yin and Detoxification on Proliferation and Cell Cycle of A549 Cells

目的观察益气化瘀养阴解毒方药物血清对人肺癌细胞A549增殖及周期的影响。方法大鼠灌胃制备益气化瘀养阴解毒方药物血清,采用MTT法观察药物血清干预24、48、72 h后对细胞增殖的作用(分为空白组及药物血清高、中、低剂量组)采用流式细胞检测分析药物血清干预24 h对细胞周期的影响(分为空白组、生长液组及药物血清高、中剂量组)。结果各药物血清组与空白组之间的吸光度差异有统计学意义(P<0.05),随着作用时间的延长,其吸光度逐渐降低,抑制率逐渐增高(P<0.05)。药物血清组细胞周期G1期增多,G2期及S期减少。结论益气化瘀养阴解毒方对肺癌细胞具有抑制作用,可能与其抑制细胞分裂有关。

Objective To observe the effect of the serum containing decoction of strengthening qi, clearing stasis, nourishing yin and detoxification on proliferation and cell cycle of A549 cells. Methods Drug- containing serum of rats were prepared. The cell proliferation after serum intervention 24, 48, 72 h was observed by MTT. The cell cycle of 24 hours intervention of the serum was analyzed by flow cytometry analysis. Results There were significant difference in absorbance between the groups of drug-containing serum and the blank group （P〈0.05）. The absorbance decreased gradually with the extension of the action time, inhibition rate increased gradually （P 〈0.05）. Cells in G1 phase increased in groups of drug-containing serum, cells inG2 phase and S phase decreased. Conclusion The decoction of strengthening qi, clearing stasis, nourishing 3,in and detoxification has inhibitory effects on lung cancer cells, may be related to the inhibition of cell division.
