摘要 目的了解转录因子E2F-1对肝癌细胞HepG2增殖的影响。方法脂质体Lipofectami-neTM2000将pCMV-E2F-1-HA2载体转染HepG2肝癌细胞,然后用含G418的培养液筛选获得具有Genectin抗性的过表达E2F-1的肝癌细胞株。通过MTT法和克隆形成实验检测细胞增殖的变化。结果 MTT和克隆形成实验结果显示,过表达E2F-1的HepG2/E2F-1细胞组与HepG2/EV细胞组和HepG2细胞组相比,其增殖速度明显减慢(P<0.01)。结论 E2F-1基因过表达可抑制HepG2肝癌细胞的增殖。 Objectlve To investigate the effects of over-expression of E2F-I on proliferation of hepato- cellular carcinoma. Methods The eukaryotic expression vector pCMV-E2F-1 -HA2 was stably transfected in- to HepG2 cell by LipofectamineTM 2000. After that, genectin-resistant E2F-1 overexpressed ceils were gath- ered by culture liquid containing O418. The effect of E2F-1 overexpression on the proliferation of HepG2 cells was observed by colony formation assay and MTT method. Results The proliferation of E2F-1 overex- pressed HepG2/E2F-1 cells was significantly lower than those of HepG2 cells and HepG2/EV cells ( P 〈 0.01 )proved by MTT and clone formation test. Conclusion Over-expression of E2F-1 gene suppresses HepG2 proliferation.
