摘要
目的探讨检测分化抑制因子1(Id1)对结肠癌HT-29细胞增殖中的影响。方法 HT-29转染Id1表达质粒及合成Id1干扰序列转染HT-29细胞后,RT-PCR和Western blot法检测VEGF mRNA和蛋白的表达,MTT法检测各组细胞增殖情况。结果 Id1过表达能显著上调VEGF mRNA和蛋白表达(P<0.05),促进细胞的生长,Id1抑制后能显著下调VEGF mRNA和蛋白表达(P<0.05),减缓细胞的生长。结论在结肠癌HT-29细胞中Id1为VEGF的上游基因,Id1可通过调控VEGF途径来影响结肠癌细胞增殖,在肿瘤血管生成中发挥重要作用。Id1有望成为结肠癌治疗的一个新靶点。
Objective To investigate the role of inhibitor of differentiation-l(Idl) on proliferation of colon carcinoma HT-29 cells. Methods The expression of VEGF mRNA and protein was detected by RT-PCR and the Western blot assay in HT-29 cells after HT-29 cells transfection with Idl expressing plasmid and Idl-siRNA. The impact on proliferation of HT-29 cells was investi- gated by MTT methods. Results The overexpression of Idl could promote both the mRNA and protein of VEGF and Idl suppres- sion could significantly reduce both the mRNA and protein of VEGF. The MTT assay showed that the cell proliferation of the sup- pression group was significantly slower than that of the control group. Conclusion Suppression of Idl can down regulate the ex- pression of VEGF in HT-29 cells and play an important role in colon carcinoma proliferation. Idl is expected to become a new target for colon cancer therapy.
