Establishment of a Highly Efficient Regeneration System from Stem Segments of‘Red Sun' Kiwifruit(Actinidia chinensis)

摘要 以"红阳"猕猴桃茎段为外植体,MS为基本培养基,培养温度25±2℃,光照强度4 500lx,16h/d,继代周期20~30d,愈伤组织暗培养.在MS+1mg/L 2,4-D+0.5mg/L 6-BA培养基中,外植体脱分化率达100%.在MS+2.0mg/L 2,4-D+0.4mg/L 6-BA培养基中,1~4代愈伤组织增殖倍数达3.38;在MS+5mg/L 6-BA+0.1mg/L NAA中,60d不定芽诱导率达83.33%.在MS+3mg/L 6-BA+1mg/L NAA中,不定芽1~6代平均繁殖系数达6.78;不定芽用0.01%IBA处理40min后在1/2MS培养基中生根率达100%;85株试管苗移栽到田间土营养钵中,15d成活81株,成活率达95.29%. Stem segments excised from one-year-old shoots of 'Red Sun' kiwifruit (Actinidia chinensis) were used as explants to induce adventitious buds on MS medium supplied with different combinations of plant growth regulators, 8 g/L agar and 30 g/L sucrose, pH=5.8, in a growth chamber set at a 16 h pho- toperiod, a 4500 lx light intensity and a 25±2℃ temperature. The calli were cultured in the dark. One hundred per cent of the explants dedifferentiated on MS+1 mg/L 2,4 D-0.5 mg/L 6 BA in a 30-day cul- ture period and the induced calli increased 3.38 times on MS+2.0 mg/L 2,4 D+0.4 mg/L 6-BA in a 20 day subculture period during 1 to 4 subcultures. Adventitious buds were regenerated from 83.33 % of the calli on MS+5 mg/L 6-BA+0.1 mg/L NAA in a 60-day culture period, and 100% of the adventitious buds multiplied and the buds proliferated about 6.78 times in every subculture during 1 to 6 subcultures on MS+3 mg/L 6-BA+1 mg/L NAA. All the adventitious buds rooted readily on 1/2 MS after treatment with 0.01% IBA for 40 minutes. Eighty-five plantlets were transplanted to a to matrix soil and, as recor- ded 15 days later, 81 were acclimatized, with a survival rate of 95.29%.
