人血清游离脂肪酸谱的气相色谱分析 被引量:3

摘要 目的 :建立用气相色谱法 ( GC)分析人血清游离脂肪酸 ( FFA)谱的方法。方法 :血清直接甲脂化 ,在 3.0 mm× 1 .0 m的不锈钢色谱柱上分离 ,正十五酸作内标。结果 :色谱分离 1 6min内完成。回收率为 84.43%~ 1 0 3.90 % ,组内和组间变异系数分别小于 5.2 8%和 7.63%。结论 :此方法不需繁杂的化学操作 ,简便迅速 ,结果准确可靠 ,适合临床使用。同时分析了正常青年组和中年组血清 FFA谱 ,两组之间差异无显著意义。 Objective: To establish a gas chromatographic (GC) method for the determination of free fatty acids (FFA) compositions in human serum Methods: A stainless steel column (3 0 mm×1 0m) packed with 15% ployethylene glycolsuccinate on chromosorb WAW DMCS is used The FFA in serum were transformed into the fatty acid methyl esters directly Pentadecanoic acid (C 15:0 ) was used as internal standard Results: The serum samples for detection and separation of the peaks for all the FFA were performed within 16 min The recoveries ranged from 84 43% to 103 90% The intra and inter assay cofficients of variaton were less than 5 28% and 7 63% respectively Conclusion: The method is simpler ,more rapid and more reliable It is suited for clinical use At the same time, we found that FFA composition of the health adult aren't significantly different between the youth and the middle age group
