硫化铅量子点对大鼠体内铜、钙、镁元素的影响

Effect of lead sulfide quantum dots on copper,calcium and magnesium in rats in vivo

摘要 目的探讨大鼠经肺染硫化铅量子点后其体内铜、钙、镁含量的变化。方法选择清洁级Wistar雄性大鼠随机分为高剂量组、低剂量组和对照组,最后经心脏取血致死,同时留取心、肝、肺、肾、脑皮质、海马和股骨标本,消化后,用Agilent 7500a型电感耦合等离子体质谱仪检测各组织的铅含量,并对相应组织中的铜、钙和镁元素含量进行测定。结果随着硫化铅量子点剂量的加大,铅染毒大鼠血液和组织中的铅元素含量显著高于对照组(P<0.05),组织中的铜、钙和镁元素含量则明显低于对照组(P<0.05);其中肝脏、肾脏和股骨为铅的主要积蓄器官,并可导致相应组织中铜、钙、镁元素含量降低,可能会影响机体的正常代谢和生理活动。结论研究提示应加强对硫化铅量子点毒性的认识,对铅暴露水平积极干预,接触人员要注意个人防护。 Objective To explore the changes of copper, calcium and magnesium in rats exposed to lead sulfide quantum dots. Methods Clean level Wistar male rats were randomly divided into three groups according to exposure dosage (high dose, low dose and normal control), at the end of experiment, all the rats were killed by drawing blood from heart, meanwhile, heart, liver, lung, kidney, cerebral cortex, hippocampus and bone tissue were also taken; then use ICP-MS (inductively Coupled Plasma Mass Spectrometry) to detect lead contents in the tissues, the levels of copper, calcium and magnesium the ac- cumulated lead were also measured at the same time. Results The results showed that with the increase of lead sulfide quantum dots exposure levels the lead contents in blood and various tissues of exposed rats were all higher than that of control group (P 〈 0. 05 ), while the copper, calcium and magnesium levels were obviously lower than that of the controls ( P 〈 0. 05 ). Conclu- sion The results suggested that the toxicity of lead sulfide quantum dots should be further realized, additionally, active inter- vention and personal protection to its exposure should also get strengthen.
