人类癫痫脑组织的蛋白质组学研究

Comparative proteomic study of epileptic foci in patients with epilepsy

摘要 目的建立癫痫患者脑组织差异表达的蛋白质谱,进一步阐明癫痫的发病机制、寻找新的治疗靶点。方法采用双向荧光差异凝胶电泳(2D-DIGE)和基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)对癫痫患者致痫灶脑组织差异表达的蛋白质进行分析鉴定。结果在患者致痫灶脑组织中共鉴定出15个蛋白质点出现1.5倍以上表达差异,其中13个下调,2个上调。结论癫痫患者脑组织中存在多个差异表达的蛋白质,可能与癫痫的发病机制相关。 Objective To construct differential express frofilings of brain tissues obtained from epilepsy patients for exploring the pathogenic mechanism and potential therapeutic targets of epilepsy. Methods Differentially expressed proteins in epileptic foci of six patients were identified by two-dimensional fluorescence difference gel electrophoresis(2D-DIGE) and matrix-assisted laser desorption/ionizartion time of flight mass spectrometry (MALDI-TOF-MS). Results 15 differentially expressed proteins which showed at least 1.5 fold difference were detected in the six patients, of which 13 were down-regulated and 2 were up-regulated. Conclusion Many differentially expressed proteins in epilepsy patients are identified, which may be involved in the pathogenesis of epilepsy.
