摘要
目的 :建立一种用反相HPLC法测定全血中环孢素A的血药浓度的分析方法。方法 :反相HPLC ,YWG -C18 柱 ,流动相为乙腈 -甲醇 -异丙醇 -水 (57∶24∶1∶18) ,流速 :1 0ml/min ,柱温 :71℃ ,检测波长212nm。结果 :环孢素A浓度在40 2μg/L~1286 4μg/L范围内有良好的线性关系 ,相关系数r=0 997。平均回收率为98 04 % ,RSD为0 6 % ,n=9。日内差RSD≤5 4 %(n=5) ,日间差RSD≤6 2 % (n=5)。结论 :此法简便、稳定 ,可应用于移植病人的CsA血药浓度监测。
OBJECTIVE:To develop a method to determine the contents of cyclosporine A in human whole blood METHO_DS:Detection was performed on YWG-C18 column with acetonitrile-methanol-isopropand-water(57∶24∶1∶18),as mobile phase Detecting wavelength was 212nm; The flow rate was 1 01ml/min and column temperature was 71℃ RESULTS:The calibration curve was linear in the concentrations ranged from 40 2μg/L to 1 286 4μg/L(r=0 997) The average recovery was 98 04% with RSD of 0 6%(n=9) The RSD within day was less than 5 4%(n=5) and the RSD between days was less than 6 2%(n=5) CONCLUSION:This method is simple and reliable and can be applied to the monitoring of blood drug concentration in patients receiving transplantation
