摘要
目的探讨TGF-α通过增加Survivin蛋白表达促进人脂肪来源干细胞(human adipose-derived stemcells,hADSCs)增殖。方法应用含不同浓度(0、0.2、0.4、0.6、1.0和2.0 ng/ml)TGF-α的培养基培养人脂肪干细胞,MTT法检测细胞的体外增殖能力,Western印迹法检测Survivin蛋白表达量。结果含TGF-α的培养基培养的人脂肪来源干细胞具有良好的细胞形态,更快的增殖速度,Survivin蛋白表达量增加,干细胞干性不受影响。结论 TGF-α通过增加Survivin蛋白表达加快人脂肪来源干细胞增殖并使其依然保持良好的干细胞特性。
Objective To investigate the effect of transformation growth factor a (TGF-a) on proliferation of human adipose-derived stem cells (hADSCs) in vitro. Methods Cultured hADSCs were treated with different concentrations of TGF-a (0, 0.2, 0.4, 0.6, 1.0 and 2.0 ng/ml). The proliferation of hADSCs was assessed by MTT method and expression of Survivin protein in hADSCs was detected by Western blotting. Results Cells in the culture medium including TGF-a had good morphology, proliferated faster and expressed more Survivin protein compared with the control group. Conclusion TGF-a enhances proliferation of hADSCs, which may be associate with up-regulation of survivin expression.
