妊娠期糖尿病胎盘组织中HIF-1α的表达及意义 被引量:5

THE EXPRESSION OF HYPOXIA-INDUCIBLE FACTOR-1α IN PLACENTA OF GESTATIONAL DIABETES

摘要 目的探讨缺氧诱导因子-1α(HIF-1α)在妊娠期糖尿病(GDM)胎盘组织中的表达及其与GDM发病的关系。方法选取妊娠36~41周行择期剖宫产的GDM孕妇44例(GDM组)和同期分娩的正常晚期妊娠孕妇37例(对照组)。采用免疫组化PV-6003两步法和逆转录-聚合酶链反应技术分别检测胎盘组织中HIF-1α蛋白和mRNA的表达。结果两组胎盘组织中均有HIF-1α蛋白的表达,GDM组HIF-1α蛋白的表达明显增强。GDM组胎盘组织HIF-1α蛋白的阳性表达率为86.4%,显著高于对照组的32.4%(χ2=22.51,P<0.05)。GDM组胎盘组织HIF-1αmRNA表达水平为0.88±0.12,显著高于对照组的0.47±0.11(t=15.91,P<0.05)。结论 GDM胎盘组织中HIF-1α蛋白和mRNA高表达可能导致胎盘组织病理形态学的异常,从而参与了GDM的发病过程。 EABSTRACT3 Objective To investigate the expression of hypoxia-inducible factor-la (HIF-la) in placenta of gestational di- abetes mellitus (GDM) and its correlation with morbility of the disease. Methods This study consisted of 44 women of 36-41 weeks of pregnancy with GDM scheduled for caesarean section (GDM group), and 37 healthy pregnant women of normal delivery (control group) of the corresponding time period. The expressions of HIF-1α protein and HIF-1α mRNA in placenta were detected by immunohistochemistry PV-6003 two-step method and RT-PCR, respectively. Results HIF-1α was expressed in the placenta of both groups, but that in the GDM group was much stronger. In GDM group, the expression rate of HIF-1α protein was 86.4%, much higher than 32.4% of that in the control (x^2 =22.51 ,P〈0.05). The expression of HIF-1α mRNA in the GDM group was 0.88±0.12 versus 0.47±0.11 of that in the control (t= 15.91 ,P〈0.05). Conclusion Overexpression of HIF-1α protein and mRNA in GDM placenta may lead to abnormal pathomorphology of placenta and invoive in the pathogenesis of GDM.
