摘要
目的:建立人结肠癌SW480细胞裸鼠皮下移植瘤模型,观察n-3 PUFA联合5-FU对SW480细胞裸鼠皮下移植瘤的疗效。方法:SW480细胞裸鼠皮下移植瘤模型随机分成正常饲料组、5-FU组、n-3 PUFA高剂量组、n-3 PUFA低剂量组、n-3 PUFA对照组,除正常饲料组、5-FU组添加正常饲料外,其余各组添加不同含量的n-3 PUFA饲料,同时,各给药组腹腔注射5-FU(35 mg/kg),一周2次,连续3周,每次给药前称体重、测量肿瘤的长径和短径;末次给药后,处死动物,剥离肿瘤并称重;通过各组肿瘤的生长曲线和抑瘤率观察SW480细胞裸鼠皮下移植瘤的抑制效果。结果:与正常饲料组比较,n-3 PUFA高、低剂量组和5-FU组肿瘤平均体积显著性减小(P<0.05);肿瘤平均重量明显减轻(P<0.05)。结论:n-3 PUFA联合5-FU对SW480细胞裸鼠皮下移植瘤有一定的抑瘤作用。
Objective:To establish the solid tumor model of human colon cancer SW480 cells, and observe the ef- ficacy in solid tumors of SW480 cells in nude mice treated with n - 3 PUFA and 5 - FU. Methods: The nude mice es- tablished subcutaneous xenograft model with SW480 cells, were randomly divided into normal diet group, 5 - FU group, n - 3 PUFA high dose group, n - 3 PUFA low dose group and n - 3 PUFA control group according to the tumor size. The normal diet group, 5 -FU group were fed with normal diet, while other groups were fed by different content of n -3 PUFA diet. At the same time, the mice in four treatment groups taken intraperitoneal injection of 5 - FU (35 mg / kg) twice a week for 3 weeks. Body weight and the diameters of tumor were measured before treatment every time. The mice were sacrificed, the tumor tissues were peeled and weighted after the last administration. The inhibitory effects of subcutaneously transplanted tumor were observed by tumor growth curves and the inhibition rate. Results: Compared with the normal diet group, the mean tumor volume in high - dose and low - dose groups of n - 3 PUFAs and 5 - FU group were significantly decreased, meanwhile average weight of tumor tissues were also significantly re- duced.
