未治疗卵巢癌中两种耐药机制的表达及其意义 被引量:1

Expression of Two Resistance Mechanisms in Untreated Human Ovarian Carcinomas

摘要 为了解卵巢癌内在性的耐药机制 ,用免疫组织化学方法对 30例正常卵巢组织及术前未经治疗的 74例卵巢癌进行了 P-糖蛋白 (P- gp)与谷胱甘肽 S-转移酶 π(GST- π)的检测 ,结果表明 :正常卵巢组织中 ,P- gp与 GST- π的阳性率分别为 1 8.9%及 74.3% ,与对照组相比 ,均有显著性差异 (P <0 .0 5 ,P <0 .0 1 )。卵巢癌中 ,单独表达一种耐药标志的有 38例 (5 1 .4% ) ,同时表达两种耐药标志的有 1 4例 (1 8.9% ) ,而且 P- gp与 GST- π的表达具有显著的相关性 (P <0 .0 1 )。结果提示 :未经治疗的卵巢癌中存在着由 P - gp与 GST- π介导的内在性的耐药机制 ,这两种耐药标志的表达 ,可能在卵巢癌的化疗耐药中起着一定的作用。 To determine the intrinsic drug resistance mechanisms of ovarian carcinomas, thirty cases of normal ovary and 74 human epithelial ovarian carcinomas of pretherapeutic patients were analyzed for the presence of P glycoprotein(P gp) and glutathione S transferase π(GST π) by means of immunohistochemistry. Results showed that P gp and GST π immunoreactivity were present in patient with ovarian caracinoma in 14(18.9%) and 55(74.3%)cases respectively, but showed negative reactions in normal ovary tissues. In the ovarian carcinomas investigated, single resistance marker was seen in 51.4% and two resistance markers in18.9% of the cases. The expression of P gp and GST π showed highly significant positive correlation (P<0.01). The results suggested that there were resistance mechanisms caused by expression P gp or/ and GST π in untreated ovarian carcinomas.The expression of the two resistance markers might contribute to drug resistance in ovarian cancers.
