摘要 目的 :应用流式细胞术 (FCM)测定血小板自身抗体 PAIg G,研究其对特发性血小板减少性紫癜 (ITP)诊断的意义。方法 :应用 FCM检测 2 0例 ITP患者血小板膜表面及血浆中的抗血小板抗体 ,并与 EL ISA所测结果进行比较。结果 :FCM测得 14例 (70 % ) ITP患者血小板膜自身抗体 PAIg G阳性 ,9例 (4 5 % )血浆抗血小板抗体阳性。EL ISA显示 12例 (6 0 % )患者 PAIg G阳性 ,7例 (35 % )血浆抗血小板抗体阳性。FCM所测结果阳性率稍高于 EL ISA。结论 :FCM可成为血小板自身抗体检测的一项新的快速简便的方法 ,与 EL ISA等方法同时应用可进一步提高 ITP患者抗血小板抗体检测的阳性率与可靠性。 Objective:Set up a method of measuring platelet associated antibody PAIgG by flowcytometry and investigate its significance in the diagnosis of idiopathic thrombocytopenic purpura (ITP).Method:With flowcytometry, PAIgG on the platelet membrane and in plasma were measured in 20 patients with ITP, which were compared with the result of ELISA.Result:By flowcytometry, the PAIgG on platelet membrane was positive in 14 ITP cases(70%), but in plasma, the PAIgG was positive in 9 cases(45%).The samples of theses patients were also measured with ELISA, which idicated positive platelet PAIgG in 12 cases (60%)and positive plasma PAIgG positive in 7 cases(35%). Compared with the results of ELISA, the positive percentage of PAIgG measured by FCM in ITP patients seems higher.Conclusion:FCM can be used as a new, rapid and simple method in detecting platelet autoantibody. FCM, in combination with ELISA, would improve the reliability and the positive percentage of PAIgG measurement in ITP patients.
