摘要
目的研究转化生长因子(TGF)-β1、纤溶酶原激活物抑制因子1(Serpine1)、血管性血友病因子(vWF)、血小板第4因子(PF4)mRNA在深静脉血栓患者血白细胞和血小板中的表达变化及在血栓形成中的作用。方法将患者分为血栓形成组15例和无血栓形成组15例,另选正常对照组15例。采集患者血栓形成和不形成时相应状态的血液样本,提取血白细胞和血小板中总RNA,并反转录为cDNA,采用PCR和实时-PCR技术,检测TGF-β1、Serpine1、vWF、PF4 mRNA在各组中的表达。结果 PCR和实时-PCR的检测结果一致,TGF-β1、Serpine1、vWF、PF4 mRNA在血栓形成组中的表达明显高于无血栓形成组和正常对照组(P<0.05),而在无血栓形成组和正常对照组间则差异无统计学意义(P>0.05)。结论血白细胞和血小板中TGF-β1、Serpine1、vWF、PF4 mRNA表达上调可能在深静脉血栓形成中发挥了重要作用。
Objective To study the change of transformation growth factor-β1 (TGF-β1), serpinel, yon Willebrand (vWF) and platelet factor 4 (PF4) expression of leucocytes and platelets in patients with deep vein thrombosis (DVT) and its role in the formation of thrombosis. Methods A total of 45 patients were enrolled in this study. The patients were divided into normal control group (n = 15), thrombosis group (n = 15) and non-thrombosis group (n = 15). Blood samples were collected in the thrombosis state as well as in the non-thrombosis state in all patients, at the same time the total RNA was extracted from the leukocytes and platelets, and its reverse transcription to cDNA was conducted. By using PCR and realtime PCR technique, the expression of TGF-β1, serpinel, vWF and PF4 in each group was determined separately. The results were analyzed. Results The results obtained from PCR technique were consistent with those obtained from real-time PCR technique. The TGF-β1, serpinel, vWF and PF4 mRNA expression of leukocytes and platelets in thrombosis group was significantly higher than that in non-thrombosis group as well as in normal control group (P 〈 0.05), while no statistically significant difference in the expression existed between the non - thrombosis group and normal control group (P 〉 0.05). Conclusion The results from present study indicate that the up-regulated TGF - β1, serpinel, vWF and PF4 expression in leukocytes and platelets may play an important role in the formation of deep vein thrombosis.
