不同时间强度冷刺激对大鼠血清中炎症相关细胞因子的影响 被引量:12

Effects of different duration of cold stress on production of cytokines in serum of SPF wistar rats

摘要 将170只SPF级Wistar大鼠随机分为常温对照组及冷刺激试验组,冷刺激试验组又分为急性应激组及慢性应激组。急性冷刺激时间为3、6、12、24h,慢性冷刺激时间为3、6、9、12d。试验大鼠均于人工气候室中饲养,对照组及冷刺激试验组的饲养温度分别为(24±0.05)℃和(4±0.05)℃。经不同时间强度的冷刺激后,采用心脏采血,采集大鼠血液,并分离血清,利用Luminex xMAP技术检测大鼠血清中IL-1β、IL-6、TNF-α、IL-2、IFN-γ、IL-4、IL-10以及IP-10的含量。结果显示,冷刺激能够提高各试验组大鼠血清中IL-6的含量,急性应激组与对照组相比TNF-α、IP-10、L-2和IL-4的含量变化水平呈上调趋势,而慢性应激组TNF-α、IL-2和IL-4的含量变化呈下调趋势,IFN-γ和IP-10的含量在冷刺激9、12d时显著增强(P<0.05)。研究结果表明,冷刺激能够引起大鼠炎症相关细胞因子以及辅助性T细胞亚群的分泌,随着应激时间的延长,细胞免疫(Th1)向体液免疫(Th2)漂移,从促炎症细胞因子(特点是IL-1和TNF-α的升高)向抗炎症细胞因子(特点是伴随IL-4和IL-10的增强)转化,机体产生细胞免疫并刺激机体产生体液免疫。 The changes and correlation of correlated cytokines in serum of SPF wistar rats under different duration of cold stress and were studied,and 170 rats were randomly divided into control groups and cold stress groups. Cold stress treatment groups were divided into acute or chronic cold stress groups. The duration of acute cold stress were 3 h,6 h,12 h,and 24 h,and the duration of chronic cold stress were 3 d,6 d,9 d and 12 d. Tenvironment temperature in cold stress groups was regulated through the artificial intelligent climate chamber The temperature of cold stress was (4±0.05)℃. After the different duration of cold stress,4 mL blood were taken from heart, and then separate serum. The levels of IL-1β, IL-6, TNF-α, IFN-γ, IL-2, IL-4, IL-10 and IP-10 were measured by Luminex xMAP technology. The results showed that the effects of different duration of cold stress on IL-1β, IL-6, TNF-a, IFN-γ, IL-2, IL-4, IL-10 and IP-10 were obviously different between control groups and cold stress groups. Cold stress enhanced pro-inflammatory cytokine IL-6 levels of all experimental groups. Acute cold stress up regulated TNF-α,IP-10 ,IL-2 and IL-4 levels. TNF-α,IL-2 and IL-4 levels showed down-regulate trends in the duration of chro-nic cold stress compared with the acute cold stress groups. Both IFN-y and IP-10 levels increased significantly at 9 d and 12 d of cold stress(P^0.05). It is concluded that cold stress induces pro- inflammatory cytokines profile and T helper cell type cytokines secretion in serum of rats. With the different duration of cold stress, the immunity level was changed from cellular immunity (Thl) to humoral immunity (Th2), and from the pro-inflammatory cytokines (characterized by enhancing of IL-1 and TNF-α levels) to the anti-inflammatory cytokines (characterized by enhan- cing of IL-4 and IL-10 levels) ,the cellular adaptive immune response and to some extent,the non- specific humoral immune response was also affected in cold stress rats.
