摘要
目的:探讨胰岛素样生长因子-1(IGF-1)在卵巢上皮性肿瘤组织中的表达,观察IGF-1反义寡核苷酸对卵巢癌细胞系3AO凋亡及侵袭能力的影响。方法:免疫组织化学方法检测60例卵巢上皮性癌、20例卵巢上皮性交界性肿瘤、20例卵巢良性肿瘤及20例正常卵巢组织中IGF-1蛋白表达;将3AO分为4组,用脂质体介导方法将IGF-1反义寡核苷酸转入其中,流式细胞仪AnnexinⅤ/PI双染色法检测其凋亡,Boyden小室检测其侵袭能力。结果:IGF-1在卵巢上皮性癌组、交界性组、良性组及正常卵巢组的阳性表达率分别为61.6%、25.0%、15.0%和10.0%,上皮癌组与其他3组比较差异有统计学意义,P<0.05;后3组之间比较差异无统计学意义,P>0.05。临床期别晚组及组织分化低组IGF-1阳性表达率为67.4%和81.8%,高于临床期别早组及组织分化高组的35.7%和52.6%,差异有统计学意义,P<0.05。IGF-1的阳性表达与发病年龄、有无腹水无关,≥50岁组阳性表达率为59.8%,<50岁组阳性表达率为54.0%,P>0.05;有腹水组阳性表达率为66.0%,无腹水组阳性表达率为52.0%,P>0.05。IGF-1反义寡核苷酸组卵巢癌细胞的凋亡率为(45.5±0.1)%,与对照组的(16.5±0.4)%比较差异有统计学意义,P<0.05;IGF-1反义寡核苷酸组癌细胞穿透Matri-gel胶的细胞数目为(42±6)个,与对照组的(132±26)个比较差异有统计学意义,P<0.05。结论:IGF-1表达与卵巢上皮性肿瘤的分期、分化有关,抑制IGF-1的表达能增加卵巢癌细胞凋亡,降低其侵袭能力。
OBJECTIVE: To investigate the expression of IGF-1 in ovarian cancer and the inhibitory effects of mvhsion of ovarian cancer cell line 3AO. METHODS: Immunohistochemical staining ASOND of IGF-1 on apoptosis and invasion of ovarian cancer cell line 3AO. METHODS: Immunohistochemical stainingwas conducted to detect the following tissue samples:60 patients with epithelial ovarian cancer, 20 patients with benign ep- ithelial,20 patients with borderline tumor and 20 patients with normal ovaries. The ASODN was transfected into 3AO cells with lipofectamineTM 2000 in the form of liposome-ODN complexes. The cell apoptosis was observed by flow cytome- try with Annexin V/PI dual staining,and the cell invasion was observed by Boyden chamber method. RESULTS: The ex- pression of IGF-1 in the group of epithelial ovarian cancer(61. 6%) was significantly higher than that in borderline (25.0%) ,benign (15.0%) and normal group 10% ,and there was no significant difference among other three groups (P 〈0. 05). The expression of IGF-1 in the ovarian cancer group of advanced stage(67. 4%) and poorly differentiated (81.8%) were significantly higher than that in early stage(35.7%) and high differentiated group(52.6% ,P〈0. 05),and the expression of IGF-lwas not correlated with age of onset (59.8% in ≥50 years old group,54.0%0 in 〈50 years old group) ,and the expression of IGF-lwas also not correlated with ascites(66.0 % in ascites group, 52.0% in non-ascites group,P〉0.05). The apoptosis of 3AO cells treated with ASODN (45.5±0. 1)% was increased significantly as com- pared with that in control group (16.5±0.4)% (P〈0.05). The number of 3AO cells that penetrated the matrigel (42± 6) was decreased singificantly, as compared with that in control group(132±26, P〈0.05). CONCLUSION : The expression of IGF-1 was correlated with the the clinical stage and differentiation of ovarian cancer, and the inhibition of IGF-I expression can increase the apoptosis of ovarian cancer cell line and decrease its invasion ability.
