摘要 目的:总结microRNA与胃癌的研究现状,探讨microRNA的生物合成、作用机制、检测方法及其在胃癌发生、发展过程中的作用。方法:采用PubMed期刊全文数据库检索系统,检索2000-01-2012-06以"microRNA、胃癌"为关键词的相关文献,共得到英文文献215条。以"microRNA的生物合成、作用机制、检测方法、胃癌发生与发展、耐药性"为纳入标准,符合条件的文献共有65篇。结果:microRNA是一种内源性非编码小RNA,能通过部分或完全与靶mRNA结合的方式调节靶mRNA的翻译,最终导致靶mRNA翻译受抑制甚至导致靶mRNA降解而失去生物活性,使相应基因表达受影响。结论:microRNA能通过转录后调控影响基因的表达,最终影响胃癌的发生、发展以及癌组织耐药性等。 OBJECTIVE: To summarize the research progress of microRNA and gastric cancer,and to investigate the biosynthesis procedure and the role in gastric cancer's happening/development of microRNA. METHODS: Based on Pubmed Journal Full text Database Retrieval Systems,literatures were searched with the key words of microRNA,Stom ach arcinoma from January 2000 to January 2012, and a total of 215 articles were obtained. Totally 65 papers were slected according to the criteria of biosynthesis,mechanism of action,detection,the role in gastric cancer's bappening/de- velopment of microRNA. RESULTS: MicroRNAs are little non coding RNA that regulate gene expression by partially or completely complementary binding to their target mRNAs, resulting in translational repression of these mRNAs,even mR- NA degradation,so the relevant gene expression is influenced. CONCLUSIONS: microRNAs can affect the expression of genes by post-transcriptional regulation,and then influence the happening and development of gastric cancer, drug resist anee and su on.
