摘要
采用蛋白酶定向水解的方法制备安康鱼皮胶原蛋白多肽,确定其最佳制备工艺。结果表明:以水解度和氮回收率为指标,筛选得出胰蛋白酶和碱性蛋白酶为胶原蛋白酶解的最佳用酶;用正交试验L9(34)对两种酶的水解条件进行优化,确定其最佳水解条件分别为:pH 10.0,酶解温度50℃,加酶量5.0 g/100 g,酶解时间5 h;pH 9.5,酶解温度50℃,加酶量4.0 g/100 g,酶解时间6 h。此外,还采用胰蛋白酶和碱性蛋白酶进行了复合酶解试验,确定复合酶解的最佳水解条件为先用胰蛋白酶最适水解条件下水解5 h,再经碱性蛋白酶最适水解条件下水解6 h,所得水解液的水解度最高,达到34.22%。
Abstract The optimal process for preparing collagen peptide from anglerfish (Lophius litulon) skin by enzymolysis was ascertained. Taking the degree of hydrolysis and nitrogen recovery as the index, the trypsin and the alkaline protease were screened for further studies. The optimization of the conditions for trypsin hydrolysis was carried out using orthogonal array design L9 (34) and the optimal hydrolysis conditions were determined as follows; the pH 10.0, the temperature 50℃, the dosage of enzyme 5.0 g/100 g, and the time 5 h; those for alkaline protease were the pH 9.5, the temperature 50℃, the dosage of enzyme 4.0 g/100 g, and the time 6 h by the same method. In addition, the compound enzymolysis was applied for the hydrolysis of anglerfish skin collagen by the trypsin and the alkaline protease, and the optimum process was initial trypsin hydrolysis for 5 h followed by another 6 h of alkaline protease hydrolysis at their optimal conditions, resulting in the generation of hydrolysate with highest hydrolysis degree. UD to 34.22%.
