摘要
目的探讨黄芪总黄酮(TFA)对嗜心性柯萨奇B3病毒(CVB3m)感染心肌细胞内向整流钾电流的作用,以明确TFA降低病毒性心肌炎小鼠心律失常发生率作用机制。方法采用体外培养小鼠新生心肌细胞方法,建立柯萨奇B3病毒感染心肌细胞模型,采用全细胞膜片钳记录技术记录心室肌细胞内向整流钾电流(IK1)。结果应用黄芪总黄酮(TFA))组与柯萨奇B3病毒感染模型组相比较,IK1峰值从(-10.11±1.57)nA增加到(-12.25±1.64)nA,差异有统计学意义(P<0.05,n=6)。结论黄芪总黄酮可增加柯萨奇B3病毒感染心肌细胞内向整流钾电流(IK1)的幅值,这可能是黄芪总黄酮降低病毒性心肌炎小鼠心律失常发生率作用机制之一。
Objective To study the effect of totalflavonoids of astragalus (TFA)on the inward rectifier potassium current of the cardiomyocytes infected by coxsackievirus B3, for identifying the mechanisms of the arrhythmias induced by viral myocarditis. Methods A toxic celluar modell was estabished in vitro by adding CVB3m into the culture of neonatal mouse cardiomyocytes. Whole cell patch clamp technique was applied to record the inward rectifier potassium current of the cardiomyocytes infected by coxsackievirus B3. Results IK 1 of TFA(20 mg/kg)increased from (-10.11±1.57)) nA to(-12.25±1.64 hA, there is significant difference compared to the cardiomyocytes infected by coxsackievirus group(P〈0.05, n=6). Conclusion The above results suggested that TFA increase the inward rectifier potassium current of the cardiomyocytes infected by coxsackievirus B3 remarkably, that probably is the one of the reasons of the arrhythmias induced by viral myocarditis.
