中药甘松挥发油对HEK细胞Na_v1.5电流频率依赖性阻滞的影响 被引量:6

摘要 目的研究中药甘松挥发油对HEK细胞Nav1.5电流频率依赖性阻滞的影响。方法用Nav1.5细胞,采取膜片钳技术直接记录甘松挥发油(VONCB)对HEK细胞Nav1.5电流频率依赖性阻滞。结果 VONCB对HEK细胞表达的鼠心肌Na+通道的具有阻滞作用。5 ppm VONCB在-100 mV电压钳制下,导致20%Na+电流抑制,静息状态产生轻微阻滞;VONCB对Na+通道抑制随刺激频率增加(1~20 Hz)而增加,产生明显的Na+通道频率依赖性阻滞。结论 VONCB对HEK细胞表达的Nav1.5通道的具有频率依性阻滞作用,VONCB对Na+通道抑制随刺激频率增加(1~20 Hz)而增加。 Objective To verify the effects of Volatile Oil of Nardostachys Chinesis Batal ( VONCB ) on frequency - dependant block of Na+ channels (Navl. 5 ) in human embryonic kidney (HEK) cells. Methods This experiment employed patch- clamp technique, VONCB and Nav 1.5 in HEK. Results 1. VONCB 5ppm , at holding potential - 100mV, the interval 30s, decreed 20% amplitude of sodium current,it is resting block. 2. VONCB 5ppm showed frequency - dependant block of sodium channel ( Navl. 5) in HEK by increasing frequency from 1 to 20 Hz. Conclusion The results suggested that VONCB showed frequency - depend- ant block of Nay 1.5 in HEK by increasing frequency from 1 to 20 Hz.
