摘要
目的探讨牛大力对小鼠辐射损伤的防护作用。方法 40只小鼠随机分为阴性对照组、辐射模型组、牛大力低(5g.kg-1.d-1)、中(10g.kg-1.d-1)、高(20g.kg-1.d-1)剂量组。用5Gy60Coγ射线对除阴性对照组外的各组小鼠进行一次性全身均匀照射,制成放射损伤动物模型。牛大力剂量组小鼠在照射前4 d及照射后10 d,以相应剂量的牛大力煎煮液连续灌胃14 d,最后一次灌胃24 h后处死小鼠,取肝、肾、肺、睾丸、脾组织进行彗星实验。结果小鼠在接受60Coγ射线照射后,牛大力剂量组各组织细胞尾部DNA百分率(Tail DNA%)和尾矩(Tail Moment)与辐射模型组相比,明显降低(P<0.01),并随着牛大力的浓度增加,Tail DNA%和Tail Moment逐步减少,呈明显的量效关系。结论在一定剂量下,中药牛大力对60Coγ射线诱导的小鼠DNA损伤有不同程度的保护和修复作用。
Objective To study the protection of Radix Millettiae Speciosae on mice radiation damage. Methods The 40 mice were divided into negative control group, radiation model group, Radix Miilettiae Speciose group with low (5 g kg-1 . d-1 ) , middle ( 10 g kg-1 d-1 ) or high(20 g kg-1 d-l ) doses. In order to establish the radiation model in mice, in addition to the negative control group, the other mice were irradiated with 5Gy 60 Coγ -rays. Radix Millettiae Speciose group mice were given with Radix Millettiac Speeiose for 4 days before irradiation and then continuously administered with Radix Millettiae Speciose for 10 days after irradiation, negative control group and radiation model group were given with normal saline by garage, once a day. After the last dose 24 hours ,Mice DNA damage of liver, kidney, lung, testicular, spleen cells was detected by comet assay. Re- suits Compared with radiation model group , Tail DNA% and Tail Moment of liver, kidney, lung, testicular, spleen cells in Ra- dix Millettiae Speciose group had significantly lower ( P 〈 0.01 ) , and with the concentration of Radix Millettiae Speciosae in- creased, Tail DNA% and Tail Moment gradually reduced, its effects were dose - dependent. Conclusion Radix Millettia Speeiosa had protection on mice DNA damage induced by 60 Coγ-rays.
