摘要
目的应用蛋白质组学技术,开展中医湿热痹阻证证候学研究。方法对类风湿性关节炎湿热痹阻证(n=12)、类风湿性关节炎肝肾阴虚证(n=12)、正常对照(n=12)共三组血清,分别进行双向凝胶电泳,通过软件分别比较湿热痹阻证组和其他两组图谱,找出共同的湿热痹阻证差异蛋白质点,应用基质辅助激光解吸飞行时间质谱进行鉴定。结果对14个湿热痹阻证共同的差异蛋白点,应用质谱进行鉴定和SwissProt数据库检索,得到9个湿热痹阻证差异蛋白,涉及细胞周期调控蛋白、分泌多肽、泛素蛋白酶体系统蛋白、膜蛋白质类、运输蛋白等。结论这些蛋白具有潜在的作为类风湿性关节炎湿热痹阻证诊断、预后标志物或治疗靶点的意义,也为进一步研究湿热痹阻证实质提供了依据。
Objective Proteomic technique was applied to analyze rheumatoid arthritis (RA) patients with damp -heat arthralgia spasm syndrome. Methods Serum proteins of RA patients with damp -heat arthralgia spasm syndrome group (n = 12 ) was com- pared with that of normal control group ( n = 12 ) and RA patients with yin deficiency of liver and kidney syndrome group ( n = 12 ) respectively by two - dimensional gel electrophoresis; the common differential protein spots of damp - heat arthralgia spasm syn- drome were analyzed by mass spectrometry. Results 14 common differential protein spots were selected and 9 proteins were iden- tified by mass spectrometry,which related to cell cycle control protein, secreted polypeptide,ubiquitin proteasome system protein, integral membrane protein, transport/cargo protein, and so on. Conclusion These proteins are of potential significanee in the diag- nosis, prognostic markers, or treatment targets of RA patients of damp - heat arthralgia spasm syndrome, and also provide a basis for further studies of the essence of syndromes in traditional Chinese medicine.
