摘要 目的应用手术联合术后一次性大剂量射线照射的方法,构建肢体淋巴水肿动物模型。方法首先手术环形切除股部宽约3cm皮肤,清除淋巴结及淋巴管,使切口暴露,瘢痕愈合,术后采用一次性大剂量射线照射,观察实验动物肢体水肿体积变化,组织中微淋巴管计数变化。结果动物死亡率为10%,在成活动物中模型成功率达83%,术区瘢痕组织中微淋巴管计数明显少于正常组织,而水肿组织中微淋巴管计数明显多于正常组织,比较有统计学意义。结论本实验表明手术与放疗相结合造模成功率较高(83%),更接近于临床,更具说服力,简单方便,是一种较好的造模方式。 Objective:To establish an improved experimental model of secondary lymphedema in limb , using radical resection of lymph vessel and a dose of radiotherapy. Methods: Modelling the clinical process, traditional radical resection of lymph vessels and nodes was performed on rabbit hindlimb. And a dose of radiotherapy was given. The volume variation was measured after operation and radiotherapy. In addition, counting of micro lymph vessels was gained and ultrasonic testing was performed to distinguish deep venous thrombosis. Results: The successful ratio was 83% in animals alive with a mortality rate of 10%. The process of edema was similar to the clinical patients after radical resection of tumor and radiotherapy. The micro lymph vessels in scar tissue were far less than that in normal tissue but the micro lymph vessels in edema tissue were excessive. Conclusion: The experimental model preparation combined operation with radiotherapy is suitable. It has a high successful ratio(83% )and is more similar to clinical case such as postoperative of breast cancer.
