酶育牛黄对肉瘤S_(180)的抑制作用及免疫调节作用的初步研究 被引量:4

摘要 目的观察酶育牛黄(CBCG)在小鼠体内、体外对肉瘤S180的抑制作用和免疫调节作用。方法 (1)体内实验:建立小鼠荷瘤模型,随机分为模型组,CBCG低、中、高剂量组和环磷酰胺(CTX)组。观察CBCG的体内抗肿瘤作用及免疫调节作用。(2)体外实验:用MTT比色法检测不同浓度含药血清分别作用于小鼠肉瘤S180细胞和小鼠脾细胞24小时和48小时后光密度值(OD值),计算细胞生长抑制/增殖率。结果 (1)体内实验:各用药组均有抑制荷瘤小鼠肿瘤生长的作用(P<0.001),且呈剂量依赖。CBCG组免疫器官指数均有所提高(P<0.001),而CTX组略有降低。(2)体外实验:各用药组均有抑制肉瘤S180细胞增殖作用(P<0.001),且呈浓度依赖;不同浓度CBCG含药血清均能促进小鼠脾细胞增殖作用(P<0.001),而CTX组对脾细胞具有抑制作用。结论 CBCG在小鼠体内、体外对肉瘤S180均有显著的抑制作用且具有较好的免疫调节作用。 Objective: To observe the inhibiting and immunomodulatory effects of culculus bovis cultivated by glucuronidase (CBCG) in vivo and in vitro. Methods: (1) Vivo experiment: The tumor -bearing mice were ran- domly divided into model group, cyclophosphamide (CTX) group and three groups of CBCG in high, medium, and low dose. The anti-tumor and immunomodulatory effects of CBCG in vivo were observed. (2) Vitro experi-ment: We detected light density value (OD value) of different concentration medicated serum acting on the mice sarcoma S180 cells and spleen cells after 24 hours or 48 hours by the MTT method, and calculated the cell growth inhibition/proliferation rate. Results : ( 1 ) The vivo experiment : The treated groups had inhibiting effect of tumor growth in tumor - bearing mice ( P 〈 0. 001 ) , and showed a dose-dependent manner. The immune organ index of CBCG groups were improved (P 〈 0. 001 ), while the CTX group decreased slightly. (2) The vitro experiment: The treated groups had inhibiting effect on sarcoma Sis0 cell ( P 〈 0. 001 ) , and revealed concentration depend- ence. Five different concentrations of medicated serum of CBCG all had a promoted effect on spleen cells of mice (P 〈 0.001 ), whereas the CTX group had inhibitory effect on spleen cells. Conclusion: CBCG has significant inhibition and immunomodulatory effect on Sis0 in vivo and vitro.
