16个高丛越橘品种VcCBF基因的单核苷酸多态性(SNP)分析 被引量:4

摘要 CBF是与植物抗寒、抗旱等抗逆性相关的一种转录因子,在植物抵抗逆境过程中发挥重要作用。以16个高丛越橘品种为试材,采用直接测序法克隆VcCBF基因,共克隆到94个非重复序列,对这些序列进行单核苷酸多态性分析,测序总长度63 732 bp,共发现103个SNP,单核苷酸多态性发生频率为1/619 bp,核苷酸多态性值(Pi)为0.01274。在103个SNP中,单态突变位点为52个,简约信息位点为51个,其中双突变为50个,三态突变为1个。通过碱基转换方式的分析发现,转换共83个,颠换19个,转换与颠换的发生比率为4.37 1。通过单倍型分析发现在16个越橘品种中VcCBF基因存在5种单倍型。 CBF, an important transcription factor, was proved to be invovled in stress-responsive in plant. Total 16 highbush blueberry cultivars were used to detect single nucleotide polymorphisms (SNPs) in VcCBF gene based on sequencing. Totally, 94 non-repetitive sequences were cloned successfully. Among these sequences, 103 SNPs were identified in VcCBF gene, on the average of one SNPs every 619 bp. The nucleotide diversity value (Pi) in 16 blueberry cultivars was 0.01274. Over the 103 SNPs, 52 of them were singleton variable sites, 50 were parsimony informative sites (two variants) and one was parsimony informative site (three variants). There were 83 transitions and 19 transversions. The rate of Ti/Tv was 4.37. Five haplotypes were finally identified for the 16 blueberry variants.
