摘要
目的:以纤维蛋白胶为载体复合胰岛素样生长因子-Ⅰ,研究该复合物对大鼠成骨细胞形态、粘附、增殖和分化的影响。方法:制备纤维蛋白胶-IGF-Ⅰ复合物,按实验分组:纤维蛋白胶-IGF-Ⅰ复合物组;纤维蛋白胶组;IGF-Ⅰ组;空白对照组,与SD乳鼠颅顶骨成骨细胞共同体外培养,采用噻唑蓝比色法、碱性磷酸酶活性及骨钙素分泌检测法以比较各组成骨细胞增殖和分化的情况。结果:第1天,四组间成骨细胞增殖及碱性磷酸酶活性差异无统计学意义,纤维蛋白胶-IGF-Ⅰ复合物组骨钙素量(OC值)与IGF-Ⅰ组差异无统计学意义,但高于纤维蛋白胶组和空白对照组,差异有统计学意义(P<0.05);第3d、5d、7d,纤维蛋白胶-IGF-Ⅰ复合物组成骨细胞增殖活力、碱性磷酸酶活性、骨钙素量均高于其余各组,差异有统计学意义(P<0.05)。结论:纤维蛋白胶有强大的粘附作用,能复合生长因子IGF-Ⅰ,并能通过逐步降解缓慢释放生长因子,促进成骨细胞的增殖与分化。
Objective: To investigate the effect of the fibrin glue(FG) as a carrier compounded with IGF- I on the proliferation and differentiation of rat osteoblasts. Methods: Osteoblasts were obtained and cultured from neonatal SD rats, and the second to fourth generations were studied. There were four groups, the FG-IGF-I group, FG group, IGF-I group, and blank control group. 3-(4, 5-Dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay, alkaline phosphatase (ALP) activity and osteocalcin (OC) were evaluated after 1, 3, 5 and 7 days. Data were analyzed by one-way ANOVA (ct = 0.05) analysis. Results: On 1 day, there was no statistically significant difference in osteoblasts proliferation and ALP activity among the four groups(P〉 0.05). FG-IGF-I group had higher osteocalcin than FG group and blank control group (P〈0.05), but had no significant difference with IGF group (P〉 0.05). After 3, 5, 7days, the proliferation rate, ALP activity and osteocalcin content of.osteoblasts in FG-IGF-I group were significantly higher than those in other groups (/9〈0.05). Conclusions: It was concluded that fibrin glue (FG) compounded with IGF-I could degrade gradually and significantly accelerate the proliferation and differentiation activity of rat osteoblasts.
