水中铜绿微囊藻定量PCR检测方法的建立 被引量:1

摘要 为了建立一种检测水中铜绿微囊藻浓度的定量PCR方法,分别用SDS裂解法和DNeasy Blood&Tissue Kit试剂盒提取标准品的DNA并建立标准曲线.结果表明采用DNeasy Blood&Tissue Kit试剂盒提取的标准品DNA具有很好的重复性,经定量PCR扩增得到的标准曲线R2为0.968,可以用来检测样品中铜绿微囊藻的浓度,为建立一套长期有效的测定水中铜绿微囊藻浓度的监测体系提供了技术参考. SDS lysis method and DNeasy Blood & Tissue Kit method were compared to establish an appropriate standard curve for the detection of Microcystis aeraginosa. A high correlation standard curve of the standard solution DNA extracted by DNeasy Blood &Tissue Kit was established(R^2=0. 968),this result above provide a new method for quantitating the Microcystis in lakes.
