HPLC法测定不同采收期猫耳朵菜中叶黄素的含量

摘要 目的:测定不同采收期光果葶苈中叶黄素的含量。方法:采用高效液相色谱法,色谱柱为C18柱(4.6mm×250mm,5μm),流动相为100%甲醇,流速1mL/min,检测波长446nm,进样量10μL,柱温室温。结果叶黄素含量在1~100μg/mL范围内线性关系良好,r=0.9977;幼苗期含量为95.50mg/kg,花蕾期含量为113.95mg/kg,花期含量为62.65mg/kg,花果期含量为35.67mg/kg,而花中含量为7.12mg/kg。结论:该方法灵敏、准确、专属性强,适用于光果葶苈中叶黄素的测定;光果葶苈不同时期的叶黄素含量存在显著差异,其中花蕾期的含量最高,花中含量最少。 Object: Determination the content of lutein in Draba nemorosa var. leiocarpa of different periods. Methods: By HPLC, chromatographic column for using Cls column (4.6 mm x 250 mm, 5μ m), mobile phase for methanol. The flow rate, injection volume of the sample and the column temperature were 1 mL/min, 10 μL, room temperature respectively and the UV detection wave-length was set at 446 nm. Results: There was good linear relationship between integral value of peak area and lutein in the range of 1-100 μg/mL (r=0.9977). The content of seedling period is 95.50 mg/kg, the content of bud period is 113.95 mg/kg, The content of flowering content is 62.65 mg/kg, the content of flower fruit period is 35.67 mg/kg, and the content of flowers is 7.12 mg/kg. Conclusions: This method is sensitive, accurate and strong specificity. It could be used for determination of lutein in Draba nemorosa var. leiocarpa. It has significant differences in different periods, among which the hiahest content of bud period.
