摘要 初步探究猴头菌生长过程中基质内部碳源代谢情况,进一步了解了木质纤维素酶酶活和还原糖含量变化与猴头菌生长发育的关系,并测定猴头菌在生长的不同阶段基质内不同部位的木质纤维素酶酶活、还原糖含量和pH值。基质内不同部位木质纤维素酶酶活和还原糖含量变化趋势基本相同,但是不同部位间酶活大小和还原糖含量有明显差异,在越靠近袋口的部位表现出越低的酶活和较少的还原糖含量。漆酶在基质刚长满菌丝时和采收后酶活较高,在原基形成时,漆酶仅在靠近袋口部位出现一个活性高峰;羧甲基纤维素酶和半纤维素酶酶活在原基形成时出现一个活性小高峰,采收时酶活达到最高;还原糖在基质刚长满菌丝时含量较高,随后降低,采收前和采收时含量达高峰。基质内不同部位pH值变化较小,未对酶活产生影响。偏相关性分析显示还原糖含量与漆酶、羧甲基纤维素酶酶活呈中度相关,与半纤维素酶酶活不相关。基质内不同部位碳代谢有明显差异,可能与基质内菌丝体处于不同生理阶段和不同分化程度相关。木质纤维素酶可能通过调节还原糖含量对猴头菌生长发育产生影响,漆酶与原基形成有密切联系。 The carbon metabolism of substrates in the development of Hericium erinaceus were investigated, and the correlation between lignocellulolytic enzyme activity and development of Hericium erinaceus were further elaborated. The lignocellulolytic en- zymes, reducing sugar content and pH of different parts of the substrates in the development of Hericium erinaceus were studied. The lignocellulolytic enzymes variation trend that the enzyme activity was much lower near the top of bag was consistent with re- ducing sugar in different parts of the substrates, although the significant differences were investigated rin content of reducing sugar and activity of the enzyme. Laccase showed higher activity after the overgrowth of mycelium in the substrates as well as in the time of harvesting, the highest activity of laccase occurred in the part near the top of bag, when the primordia were forming. Carboxym- ethyl cellulose enzyme and hemicellulase also showed a higher acitivty in the primordia formation, but they displayed the summit of activity in the time of harvesting. Much more reducing sugar content appeared in the overgrowth of mycelium in the substrates with subsequently less content, and the most content occurred before or at the time of harvest. The pH value varied slightly in dif- ferent parts of the substrates, and it did not take any effect on enzyme activity. Partial correlation analysis indicated the reducing sugar had moderately correlation with laccase and carboxymethyl cellulose enzyme, and no correlation with hemicellulase. The sig- nificant differences of carbon metabolism probably correlate with the different physiology and development period of mycelium in different parts of the substrates. Lignocellulolytic enzyme had impact on the development of Hericium erinaceus by regulation of re- ducing sugar content. The primordia formation was closely related to laccase.
