摘要
目的:研究Hsp70在TLR信号途径中对肿瘤细胞增殖的影响。方法:将体外培养的H22细胞分成对照组和用Hsp70刺激的实验组,设计基因表达的引物、PCR扩增、RT-PCR检测两组细胞的TLR2和TLR4的mRNA表达水平,Western blot检测β-catenin水平,对培养10天后的H22细胞进行计数。结果:Hsp70分别处理H22细胞8h后,基因TLR2、TLR4mRNA的表达均明显上调;Hsp70促进H22细胞中β-catenin上调表达;Hsp70处理组的细胞增殖倍数高于对照组。结论:Hsp70在TLR信号途径中具有促进肿瘤细胞增殖作用。
Objective:To research the effect of Hsp70 for tumor cell proliferation in the TLR signaling pathway. Methods:h put H22 cells in vitro into a control group and the experimental group stimulated with Hsp70 and designed the gene expression primers, PCR amplification, RT PCR was used to detect the two groups of ceils TLR2 and TLR4 mRNA expression, Western blot detection of β-catenirfs level, H22 cells were cultured for 10 days were counted. Results:The expression of gene TLR2, TLR4 mRNA was increased significantly treated by Hsp70 after eight hours in H22 cells; the β-catenin was upregulated by Hsp70 promote in the H22 cells ; the proliferation multiples were higher in Hsp70-treated H22 cells than control group. Conclusion : The Hsp70 was promoted for tumor cell proliferation in the TLR signaling pathway.
