消岩汤对肺癌恶病质小鼠肿瘤生长的影响 被引量:17

Effect of Xiaoyan Decoction on Cachexia and Tumor Growth in Mice with Lung Cancer

摘要 目的研究消岩汤对肺癌恶病质小鼠肿瘤生长的影响,并探讨其可能作用机制。方法将40只小鼠分为健康小鼠组(健康组)、Lewis肺癌小鼠组(肺癌组)、Lewis肺癌小鼠消岩汤组(消岩汤组)、Lewis肺癌小鼠醋酸甲地孕酮组(孕酮组),每组10只。连续给药12d后,测定血清瘦素水平及瘤组织中p53分子表达及C-myc蛋白表达。结果除健康组外,消岩汤组、孕酮组小鼠反应性及体毛较肺癌组明显改善,进食量明显增加(P<0.01),其中消岩汤组进食量多于孕酮组(P<0.05)。肺癌组小鼠反应性及体毛情况恶化最明显。3组进食量均低于健康组(P<0.01)。给药后处死各组小鼠,肺癌组、消岩汤组、孕酮组小鼠体重均比健康组小鼠体重减轻(P<0.05或P<0.01)。至给药第12天,消岩汤组、孕酮组瘤重明显较肺癌组轻(P<0.01),消岩汤组瘤重低于孕酮组(P<0.01)。肺癌组、消岩汤组、孕酮组血清瘦素水平均低于健康组,其中肺癌组、孕酮组与健康组比较下降明显(P<0.05);而孕酮组低于消岩汤组(P<0.05)。消岩汤组、孕酮组的p53及C-myc蛋白表达水平与肺癌组比较差异均有统计学意义(P<0.01),消岩汤组与孕酮组比较差异亦有统计学意义(P<0.01)。结论中药消岩汤具有增加恶病质小鼠食量和体重、改善机体机能、升高血清中瘦素受体水平、促进p53的表达并抑制C-myc表达的作用,达到抑制肿瘤生长的作用。 Objective To observe the effect of Xiaoyan Decoction (antitumor decoction) on cachexia in mice with lung cancer and its antitumor mechanism. Methods Forty mice were randomized into the healthy group, lung cancer group, Xiaoyan De- coction group and megestrol acetate group, with 10 in each. The leptin content and the expressions of p53 and C-myc protein were observed after 12 days of continuous treatment. Results Comparing with the lung cancer group, tb.e reaction, body b.air and food intake improved significantly in the Xiaoyan Decoction group and megestrol acetate group (P〈0.01), especially the food intake in the Xiaoyan Decoction group (P〈0.01). The situation of reaction and body hair was the worst in the lung cancer group. The mice were sacrificed after treatment. Comparing with the healthy group, the bodyweights in the lung cancer group, Xiaoyan Decoction group and megestrol acetate group were significantly decreased (P〈0.05 or P〈0.01), especially in the lung cancer group (P〈0.01), especially in the Xiaoyan Decoction group (P〈0.01), Comparing with the lung cancer group, the tumor weight was significantly decreased in the Xiaoyan Decoction group and megestrol acetate group on the 12th day of treatment (P〈0.01), especially in the Xiaoyan Decoction group (P〈0.01). Comparing with the healthy group, the leptin levels in the lung cancer group, Xiaoyan Decoction group and megestrol acetate group were decreased, especially in the lung cancer group and megestrol acetate group (P〈0.05). Comparing with the lung cancer group, the expressions of p53 were sig- nificantly increased in the Xiaoyan Decoction group and megestrol acetate group (P〈0.01), especially in the Xiaoyan Decoction group (P〈0.01). Comparing with the lung cancer group, the expressions of C-mye were significantly decreased in the Xiaoyan Decoction group and megestrol acetate group (P〈0.01), especially in the Xiaoyan Decoction group (P〈0. 01). Conclusion Xiaoyan Decoction can improve appetite, body function and immune function, increase bodyweight and leptin level, promote p53 expression and inhibit C-myc expression and tumor growth in mice with lung cancer.
