分光光度法测定锦鸡儿中的总黄酮

Determination of Total Flavonoids in Caragana sinica by Spectrophotometry

建立锦鸡儿中总黄酮的含量测定方法和最佳提取工艺。采用正交试验设计,以总黄酮含量为考察指标,研究锦鸡儿总黄酮的最佳提取方法;并利用分光光度法,以芦丁为对照品,对产于安徽的锦鸡儿原药材中的总黄酮含量进行测定。该药材总黄酮的含量为3.37%;最佳提取工艺为以90%的乙醇为溶剂、物料比1:15g/mL、回流提取3次,每次60min。该工艺合理,黄酮有效成分提取率较高,可作为锦鸡儿中黄酮的提取方法。

Method for determining of total flavonoids in Caragana sinica and the optimum extraction conditions were developed. With the content of total flavonoids as index, the optimum extraction method of total flavonoids in Caragana sinica was investigated by orthogonal test design, and the content of total flavonoids in Caragana sinica original herbs from Anhui was determined by spectrophotometry with turin as standard sample. The results showed that the average content of total flavonoids in the herbs was 3.37%. The optimum extraction conditions were as follows. 90% ethanol as the solvent,the liquid-solid ratio of 1：15g/mL,reflux extraction of three times with 60min each time. The method is reliable with high extraction rate,and can be used in the extraction of flavonoid in Caragana sinica.
