摘要
研究蓝莓总黄酮的大孔树脂纯化工艺及其抗氧化活性。以静态饱和吸附量和解析率为指标,对4种大孔树脂(AB-8、NKA-9、NKA-Ⅱ、D101)进行筛选;以回收率为指标,通过选用L_9(3~4)正交表设计实验,确立纯化总黄酮的最佳条件。以VC和芦丁为对照品,考察蓝莓总黄酮对DPPH自由基(DPPH·)和羟基自由基(·OH)的抗氧化活性。结果表明NKA-Ⅱ纯化蓝莓总黄酮效果最好,最佳纯化工艺为:上样浓度1.355mg.mL^(-1),洗脱pH为3,洗脱乙醇浓度70%,洗脱流速2mL.min^(-1)。在此条件下纯化的总黄酮纯度可达41.41%,比未纯化前提高33.94倍。蓝莓总黄酮对DPPH·和·OH的抗氧化活性以半数抑制浓度(IC_(50))表示分别为0.0217和0.1037mg.mL^(-1)。NKA-Ⅱ大孔树脂综合性能较好,适于分离纯化蓝莓总黄酮,蓝莓总黄酮具有较强的抗氧化活性。
-To study the fiavonoids purification by macroporous resin and antioxidant activity of flavonoids from blueberry.Four types of macroporous resin (AB-8, NKA-9, NKA- Ⅱ, D101 ) were used to purify flavonoids with static adsorption capacity and elution ratio as evaluation factors .Then the optimum purification conditions were investigated by L9 (34) orthogonal experimental design with recovery as evaluation factor.The antioxidant activity of the fiavonoids from blueberry was tested by scavenging DPPH ~ and ~ OH with Vc and lutin as references. NKA-]][ was the most suitable type of macroporous resin and the optimum conditions were as follows: the concentration of sample was 1.355mg ~ mL 1, the washing pH was 3, the flow rate of 70% ethanol was at 2mL ~ min-1, in this condition the purity of total flavonoids could reach 4].41% ,33.94 times than before.The IC50 of antioxidant activity were 0.0217 and 0.1037mg· mL-1 respectively. Conclution was that the NKA-1Ⅱ type macroporous resin showed better comprehensive absorption property,it can be used to purify the fiavonoids from blueberry,the flavonoids from blueberry has strong antioxidant activity.
