摘要
目的观察针刺督脉经穴对脑缺血再灌注大鼠缺血半暗带神经细胞骨架结构微管相关蛋白MAP-2和神经微丝蛋白NF-L的mRNA表达的影响,并探讨其可能的作用机制。方法以针刺督脉经穴为干预手段,以大鼠急性局灶性脑缺血再灌注模型作为研究对象,运用SYBR Green实时定量PCR观测脑缺血再灌注后缺血半暗带MAP-2和NF-L的mRNA表达变化,并观察针刺对上述指标的调节作用规律。结果针刺组和对照组MAP-2和NF-L的mRNA表达较模型组均显著升高(P<0.01),且针刺组升高更显著(P<0.05)。结论针刺督脉经穴能上调MAP-2和NF-L的mRNA表达,促进神经元树突和轴突的出芽和再生,提高神经元的可塑性,可能是针刺督脉经穴发挥神经保护作用的机制之一。
Objective To investigate the effect of acupuncture at Du meridian points on the expressions of neuronal microtubuleassociated protein 2 (MAP-2) mRNA and neurofilament protein L (NF-L) mRNA in the ischemic penumbra in rats with cerebral ischemia and reperfusion and explore the possible mechanism of its action. Methods Acupuncture at Du meridian points was used as an Interventional measure. A rat model of focal cerebral ischemia and reperfusion was used as a subject. The expressions of MAP-2 and NF-L mRNAs in the ischemic penumbra were examined using SYBR Green real-time and quantitative PCR after cerebral ischemia and reperfusion. Regularities in the adjusting effect of acupuncture on the above indices were observed. Results The expressions of MAP-2 and NF-L mRNAs increased significantly in the acupuncture and control groups compared with the model group (P〉0.01) and increased more in the acupuncture and control group (P〈0.05). Conclusion Acupuncture at Du meridian points can upregulate the expressions of MAP-2 and NF-L mRNAs, promote the germination and regeneration of neuronal dendrites and axons and improve neuronal plasticity. That may be one of the mechanisms by which acupuncture at Du meridian points produces a protective effect on the nerve.
