青霉α-半乳糖苷酶基因工程菌表达条件优化

摘要 试验通过测定基因工程菌的酶活性,研究了培养基pH、摇床转速、接种量和甲醇浓度对青霉α-半乳糖苷酶表达的影响,旨在优化重组菌株的表达条件。结果表明:当培养基pH为5,摇床转速为200 r/min,种子接种量为5%,甲醇浓度为1.0%时,发酵上清液酶活性最高。 To optimize the expression conditions of the recombinant PeniciUium a-galactosidase,the effects of the culture pH value,the speed of the shaker,inoculum sizes and the induced methanol concentration were investigated through measuring the activity of the enzyme in this paper.The results showed that:with the culture pH value 5 ,the speed of the shaker 200 r/min,inoculum sizes 5%, and the induced methanol concentration 1.0%,the activity of the fermentation supernatant was the highest.
