鞘内注射右美托咪定对慢性炎性痛小鼠的影响 被引量:2

摘要 目的探讨鞘内注射右美托咪定(dexmedetomidine,Dex)对慢性炎性痛小鼠的影响。方法雄性昆明小鼠随机分为对照组(C组:足底注射30μL生理盐水)、慢性炎性痛组(P组:足底注射30μL完全弗氏佐剂)、Dex组(D组:鞘内注射Dex 1μg)和Dex定+慢性炎性痛组(D+P组:足底注射30μL完全弗氏佐剂3 d后鞘内注射Dex 1μg),鞘内注射Dex前,注射后30、60、90、120、150、180、210 min(T0~T7)检测热缩足潜伏期(n=8)和机械缩足阈值(n=8),在T4时间点行免疫组织化学法检测脊髓Fos蛋白表达并计数(n=6)。取正常小鼠18只,随机分为D0、D1和D3组,D0组不做处理,D1和D3组鞘内注射Dex(1μg/次,每12小时1次),分别持续1 d和3d,末次给药后12 h取脊髓行HE染色。结果 T0~T5时间段,与C组比较,P组热缩足潜伏期和机械缩足阈值明显降低(P<0.05),T1~T5时间段,D组和D+P组较P组明显升高(P<0.05)。在T4时间点,与C组比较,P组脊髓背角Fos蛋白表达明显增加(P<0.05),D组和D+P组较P组明显减少(P<0.05)。D1、D3组与D0组比较脊髓无明显病理变化。结论鞘内注射Dex对慢性炎性痛具有镇痛作用,且对脊髓无明显病理损伤。
