摘要
目的观察含Ⅰ型血小板结合蛋白基序的解聚蛋白样金属蛋白酶(ADAMTS-1)在实验性肺纤维化大鼠血清中的变化。方法 50只SD大鼠随机分为对照组(n=10)、模型组(n=40),模型组经气管内注入博莱霉素建立大鼠肺纤维化模型,对照组相同条件下给予生理盐水,模型组分别于第7、14、28、56天随机处死10只大鼠,而对照组于第56天处死,取出肺组织行HE染色和Masson染色,采用酶联免疫吸附法检测血清ADAMTS-1水平。结果模型组各时间点血清ADAMTS-1水平显著低于对照组(P<0.01),模型组血清ADAMTS-1水平与肺纤维化程度评分呈负相关(r=0.74,P<0.05)。结论 ADAMTS-1可能参与肺纤维化发病过程,并可作为病情判断的观察指标。
Objective To observe the changes of a disintegrin-like and metalloproteinase with thrombospondin type I motif ( ADAMTS-1 ) in experimental rats with pulmonary fibrosis. Methods 50 SD rats were randomly divided into the control group ( n = 10) and the model group ( n = 40). The model group were injected with bleomycin through trachea to produce pulmonary fibrosis model, and the control group were treated with normal saline at the same condition. 10 rats in the model group were killed on the 7th, 14th, 28th and 56th days while all rats in the control group were killed on the 56th day. Then the pulmonary tissues were removed and given HE staining and Masson staining. The level of serum ADAMTS-1 was examined by ELISA. Results The level of serum ADAMTS-1 at every time point in the model group was lower than that in the control group (P 〈0. 01 ). The level of serum ADAMTS-1 was negatively eorrelatod with the score of pulmonary fibrosis degree in the model group ( r = 0. 74, P 〈 0. 05 ). Conclusion ADAMTS-1 may take part in pulmo- nary fibrosis, and it can be used as an indicator for pathogenetie condition.
