欣胃颗粒对胃癌前病变大鼠STAT3、p-STAT3信号通路表达影响的研究

Effects of Xinwei Granules on STAT3 and p-STAT3 Singal Pathway in Rats with Precancerous Lesions of Gastric Carcinoma

目的:探讨欣胃颗粒对胃癌前病变(PLGC)大鼠STAT3、p-STAT3信号通路的影响。方法:采用N-甲基-N'-硝基-亚硝基胍(MNNG)为主的复合因素造模法制备PLGC大鼠模型,造模成功后,随机分为6组,模型组、欣胃颗粒高、中、低剂量(高、中、低剂量)组,维酶素对照(维酶素)组,另设空白对照(空白)组未予造模。各组分别给予相应的处理,12周实验结束,观察各组大鼠的一般状态和胃黏膜组织病理学的变化,观察其对STAT3、p-STAT3表达的影响。结果:欣胃颗粒各剂量组大鼠活动度、进食水量、体重等一般状态好转;模型组与空白组比较,STAT3、p-STAT3表达水平增高,欣胃颗粒低、高剂量组与模型组比较,STAT3、p-STAT3表达水平降低,差异均有统计学意义(P<0.05)。结论:欣胃颗粒对PLGC大鼠胃黏膜组织病理具有明显的改善作用,可以下调STAT3、p-STAT3的表达以达到对抗PLGC的作用。

Objective：To study the effect o f Xinwei Granules on STAT3 and p - STAT3 singal pathway in rats with precancerous lesions of gastric carcinoma. Methods ： The rat model of PLGC established by complex pathogenic factor in which MNNG was mainly used. All of the rats were randomly divided into five groups, including blank group, model group,Xinwei Granules on iow- dose and high - dose group, Vitacoenzyme group and normal control group. After 12 weeks,the general conditions, the pathological changes in rats gastric mucosa as well as STAT3 mRNA and p -STAT3 expressions were observed. Results：General conditions, such as activity eating and body weight were improved in all Xinwei Granules groups ; compared with the blank group, the STAT3 and p - STAT3 expressions of in the model group were significantly increased, the differences between these two groups were statistically significant （P 〈 0.05 ） ;compared with that of model group,the expressions of STAT3 and p- STAT3 in different doses of Xinwei Granules groups were significantly re- duced, the differences were statistically significant （ P 〈 0. 05, P 〈 0.01 ）. Conclusion ： Xinwei Granules groups can im- i3rove the pathological conditions of gastric mucosa in rats with PLGC ; and it also can prevent the progress of PLGC by re- ducing Stat3 mRNA expression and p- stat3 levels.
