摘要
目的:观察不同浓度的鸡肉、鸭肉提取液分别对体外培养人肺腺癌A549细胞血管内皮生长因子(vascular endothelial growth factor,VEGF)及DNA合成的干预作用,探讨鸡肉、鸭肉提取液对体外培养人肺腺癌A549细胞影响的作用机制,以期为临床肿瘤患者合理饮食提供指导。方法:体外培养人肺腺癌A549细胞株,细胞生长至对数生长期后,随机分为9组进行培养。采用VEGF-ELISA法检测肺腺癌A549细胞上清液中的VEGF浓度,BrdU-ELISA法检测肺腺癌A549细胞DNA的合成情况。结果:与对照组相比,鸡肉提取液、鸭肉提取液均能促进肺腺癌A549细胞血管内皮生长因子的生长,无统计学差异(P>0.05)。鸡肉提取液、鸭肉提取液均能够促进肺腺癌A549细胞DNA的合成,差异有统计学意义(P<0.01),且随着剂量的增加DNA的合成越多。结论:鸡肉、鸭肉提取液并没有通过促进肺腺癌A549血管内皮生长因子生长来促进肿瘤细胞生长。鸡肉、鸭肉提取液均能够促进肺腺癌A549细胞DNA的合成,使细胞的DNA合成期(S Phase;S期)增多,S期增多可能是其促进细胞增殖的机制之一。
Objective:To provide better instructions of healthy diets for cancer patients, the influence of chicken or duck meat extracts on human lung adenocarcinoma A549 cells was studied. The proliferation or inhibition of lung adenocarcinoma A549 cells was investigated by monitoring the vascular endothelial growth factor(VEGF) and the synthesis of DNA. Method:lung adenocarcinoma A549 cells cultured in vitro grew to logarithmic phase, then they were divided into nine groups and fed with different concentrations of chicken or duck meat extracts. The concentration of VEGF in supernatant was quantified by VEGF - ELISA and the DNA synthesis of A549 cells was quantified by Brdu - ELISA. Results : Chicken and duck meat extracts slightly promote the growth of VEGF in A549 cells but without statistical significance compared with control groups (P 〉 0.05 ). However, chicken and duck meat extracts significantly promote the DNA synthesis of lung adenocarcinoma A549 cells and the DNA amounts were increased with the additional doses of meat extracts (P 〈0.01 ). Conclusions: Chicken and duck meat extracts did not promote tumor cell growth through promoting the growth of VEGF of lung adenocarcinoma A549. Instead, they promoted the DNA synthesis and increased cells in S period, which may be one of the mechanisms that promote cell proliferation.
