摘要
目的:以免疫组化技术观察糖尿病大鼠坐骨神经中转录因子IκBa、Ikka的蛋白表达。方法:148只雄性Wistar大鼠,以STZ诱导糖尿病模型,随机分为正常组、模型组、辛味通络组、藤类通络组、虫类通络组。于治疗后4、8周处死大鼠取坐骨神经备免疫组化观察用。结果:造模组IκBa、Ikka水平明显低于同期正常组(P<0.01),与同期模型组比较,通络中药组均能提高IκBa、Ikka的表达(P<0.01),同期治疗组之间无显著性差异。结论:不同通络中药随着疗程延长均具有延缓神经病变进程的作用,可能与调节IκBa、Ikka信号系统有关。
Objective : To observe the expressions of transcription factors IKBa and Ikka in the sciatic nerve of diabetic rats by immunohistochemical technique. Method: 148 male Wistar rats were randomly divided into five groups:normal- control group ( NC ), model - control group ( MC ), Xinwei Tongluo group ( XW ), Tenglei Tongluo group ( TL ), Conglei Tongluo group (CL), and diabetic model was induced by STZ, then the rats were killed in 4th and 8th week, and the sciatic nerve was taken out and tested by immunohistochemical method. Results : The levels of IκBa and Ikka of MC were obviously lower than N C in the corresponding time period (P 〈 0.01 ), compared with MC, the different smoothing collaterals groups can elevate the expressions of IκBa and Ikka(P 〈0. O1 ). There was no obvious difference during the three therapeutic groups at the same time. Conclusion : The different collaterals - smoothing drugs can lessen the damages to structure and function of peripheral nerves in diabetic rats and postpone the development of neuropathy. This protective effects are possibly related with regulating the IKBa and Ikka signal transduction system.
