摘要
目的:观察血府逐瘀汤(XFZYT)对野百合碱(MCT)诱导肺心病大鼠肺血管重构的干预作用,探讨其作用机制。方法:雄性SD大鼠54只,随机均分为正常对照组(Control)、模型组(Model)、血府逐瘀汤预防组(XFZYT1)、血府逐瘀汤治疗组(XFZYT2)、XFZYT1组的硝苯地平对照组(Nifedipine1)和XFZYT2组的硝苯地平对照组(Nifedipine2)。采用一次性腹腔注射MCT(60 mg/kg)复制肺血管重构大鼠模型。药物干预后测定各组大鼠肺血管病理形态学变化,并采用免疫组化技术比较各组大鼠肺血管增殖细胞核抗原(PCNA)及Caspase-3蛋白表达情况。结果:与模型组比较,各药物组均能使肺小动脉管壁有不同程度变薄、管腔变大(P<0.01或P<0.05);血管增殖细胞核抗原(PCNA)增殖度下降,Caspase-3蛋白阳性率升高(P<0.01或P<0.05)。结论:血府逐瘀汤能有效减轻肺心病大鼠肺血管重构,其机制可能通过调节平滑肌细胞增殖与凋亡有关。
Objective:To observe the effects of Xuefu Zhuyu Decoction on remodeling of pulmonary vascular in chronic col pulmonale rats induced by monocrotaline.And further explore the possible mechanism to provide theoretical basis for its further application in clinic.Methods:Male SD rats were randomly divided into normal control group,model group,Xuefu Zhuyu Decoction prevention group,Xuefu Zhuyu Decoction treatment group,nifedipine prevention group and nifedipine treatment group(n=9 for each group).Rats were given a single dose of MCT(60 mg/kg)to induce the model of chronic col pulmonale.The pathological features of pulmonary arteries were observed by microscope after intervention of drugs.Immunohistochemical stains were used to identify proliferation cells nuclear antibody(PCNA)and Caspase-3 protein distribution in pulmonary arteries.Results:PCNA expression level in each drug group was lower than that in model group,while Caspase-3 in each drug group was significantly higher(P〈0.01 or P〈0.05).Conclusion:Xuefu Zhuyu Decoction might reduce the remodeling of pulmonary vascular by adjusting the expressions of PCNA and Caspase-3 protein.
